Etoricoxib belongs to the coxib class of NSAIDs and works by selectively inhibiting the cyclooxygenase-2 (COX-2) enzyme. COX-2 is induced at sites of inflammation and is primarily responsible for the synthesis of prostaglandins that mediate pain, inflammation, and fever. By blocking COX-2, Etoricoxib reduces the production of these inflammatory mediators, providing effective relief from pain, joint stiffness, and swelling.
Importantly, at therapeutic doses, Etoricoxib has minimal effect on the cyclooxygenase-1 (COX-1) enzyme. COX-1 is constitutively expressed in the stomach, kidneys, and platelets, where it produces prostaglandins that protect the gastric mucosa, maintain renal blood flow, and support normal platelet function.
